[STEMvision™]
STEMvision™ is an automated instrument and computer system
designed for imaging and scoring hematopoietic colonies
in the colony-forming cell or CFC assay.

STEMvision™ reduces variability in CFC assay results,
saves time, and acquires digital images for permanent record keeping.
Instead of manually identifying and counting colonies using a microscope,
users simply load cultures into STEMvision™.
The instrument automatically images and analyzes cultures
using highly sophisticated software and colony-counting algorithms.
STEMvision™ is optimized for use with MethoCult™ media
and new meniscus-reducing SmartDish™ cultureware.

STEMvision™ increases standardization of colony scoring
reducing any variability associated with scoring colonies,
both between individuals within a laboratory
and between different laboratories.
STEMvision's™ sophisticated colony identification algorithms
ensure the same criteria for colony identification and classification
are consistently applied independent of the operator or laboratory.
Comparing the 95% confidence intervals
of automated colony counts using STEMvision™ and manual counts
demonstrates the significant reduction in variability.
Results from the CFC assay obtained using STEMvision™
correlate strongly with manual counts.
This graph shows the correlation between manual and automated counts
for the 7-day assay using MethoCult™ Express.
Similar results have been obtained for the 14-day assay
using MethoCult® H4034 Optimum.

STEMvision™ offers laboratories a significant time savings.
Imaging each SmartDish™ well
is accomplished in approximately 30 seconds,
while scoring analysis for each well is completed in approximately 3 minutes.
The STEMvision™ system allows high-throughput processing
for labs that handle a large number of samples.
For example, users can image dishes one after the other
with twenty 6-well SmartDishes™ containing 120 cultures
requiring approximately 1 hour.
The scoring analysis can then be completed in an overnight step.

Driven by STEMvision's™ software package,
high-resolution images are acquired at 3.4 microns per pixel.
Using the analysis function colonies will be identified on the image
and available for review.
STEMvision's™ imaging function is useful for time-lapse studies.
The ability to record and review colony proliferation and differentiation over time
allows for detailed characterization studies.

STEMvision™ has been optimized for use with MethoCult™,
the gold standard methylcellulose-based medium for
CFC assays and SmartDishes™,
meniscus-free cultureware developed by Stemcell Technologies
for greater colony counting accuracy.
MethoCult™ Express is formulated for accelerated
progenitor proliferation and colony formation
allowing for enumeration of total progenitors after only 7 days
without distinguishing different colony types.
Stemcell also offers MethoCult™ media products
appropriate for the 14-day CFC assay.
For both the 7- and 14-day CFC assays,
the use of SmartDish™ cultureware will ensure consistent
and accurate colony counting results.
SmartDishes™ reduce the formation of meniscus
permitting more uniform distribution of the medium.
The result is more homogeneous colony distribution
and less optical distortion and crowding of colonies at the edge of the dish.
This is in contrast to traditional cultureware
where the meniscus effect leads to greater medium depth
and proportionally higher colony numbers at the edge of the culture dish
where they are also more difficult to identify.
