here's a look at what's coming up in the may two thousand thirteen issued of JoVE
the journal visualized experiments
what came first the chicken or the egg
this fundamental question is missing a final component of reproduction
the ***
before anything can happen a *** cell must swim to an egg and fuse with it
these events depend a lot on signal transduction pathways mediated by
calcium ions
therefore to study *** function it's useful to measure changes and calcium
ion concentration in spermatozoa
this can be facilitated by calcium sensitive fluorescents dyes
which our authors use and four fluora metric techniques to monitor calcium
dynamics in human ***
conventional flowometry
stopped flow flowometry
flow cytometry and single cell imaging
each technique can allow researchers to study the role of calcium ion
fluctuation and spermatozoa or other cells
moving along to another part of the male reproductive tract we feature an article
concerning prostate cancer one of the leading causes of cancer deaths in
american men
our office recently showed that an enzyme called argentine deaminase or
ADI
causes human prostate cancer cells to undergo atophagy
a mechanism of cell death in which a cell essentially eats its own components
to study this mechanism our authors developed an imaging based approach
using quantitative three-d_ fluorescence microscopy
this allows them to precisely track morphological changes as the cells
undergo atophagy
this technique will help researchers study potential therapeutics for
prostate cancer
in JoVE bioengineering
past articles have presented methods for isolating silk producing glands from
spiders producing recombinant spider silk proteins and bacteria
and spinning purified proteins into fibers potential biomedical applications
this month we feature a new application for spider silk
making air filter devices with a non-woven mess of recombinant spider
silk proteins
as we all know spider silk is extremely strong and very sticky
therefore if commercial filters coated with a layer of spider silk
their filtering efficiency may be improved
our authors demonstrate how to electrospin in the fibers treat the non-woven self
meshes and analyze the meshes using scanning electron microscopy
finally our authors demonstrate air permeability and filter efficiency
in JoVE medicine we feature a method for determining the lowest dose of
ultraviolet light that'll cause erythema
or sunburn in
an individual
u_v_ light is often used to treat various skin conditions such as cirrhosis, acne
and eczema
because not all people equally sensitive to u_v_ light this method can help
determine the right dose to use
this procedure uses at daavlin patch to control both the area and duration of
u_v_ exposure on the skin
the next day changes in skin color are assessed to determine the lowest UV
dose required to cause erythema
finally in JoVE neuroscience we have an article demonstrating how to
culture slices of embryonic mouse neuro epithelium and performed live imaging of
various fluorescence markers
this method allows researchers to monitors cell behavior
such a single cell divisions in situ and in real-time
